Composition

active ingredient : dexketoprofen;

1 film-coated tablet contains dexketoprofen 25 mg in the form of dexketoprofen trometamol;

excipients : microcrystalline cellulose (pH 101), corn starch, sodium starch glycolate (type A), glycerol distearate.

Shell: Opadry II 85F18422 white (polyvinyl alcohol, titanium dioxide (E 171), polyethylene glycol, talc).

Pharmacodynamics

Dexketoprofen trometamol is the tromethamine salt of S-(+)-2-(3-benzoylphenyl)propionic acid, which is an analgesic, anti-inflammatory and antipyretic drug that belongs to the group of non-steroidal anti-inflammatory NSAIDs.

The mechanism of its action is based on a decrease in the synthesis of prostaglandins due to the inhibition of cyclooxygenase. In particular, the conversion of arachidonic acid into cyclic endoperoxides PGG 2 and PGH 2 is suppressed , from which prostaglandins PGE 1 , PGE 2 , PGF 2a , PGD 2 are formed , as well as prostacyclin PGI 2 and thromboxanes (TxA 2   and TxB 2 ). In addition, inhibition of prostaglandin synthesis may affect other inflammatory mediators, such as kinins, which may also indirectly affect the main effect of the drug.

The inhibitory effect of dexketoprofen trometamol on cyclooxygenase 1 (COX-1) and cyclooxygenase (COX-2) activity has been demonstrated in animals and humans.

In clinical studies conducted on several models of pain, the effective analgesic activity of dexketoprofen trometamol has been proven. The onset of analgesic action is achieved 30 minutes after the use of the drug and lasts 4-6 hours.

Indications

Symptomatic treatment of mild to moderate pain, such as musculoskeletal pain, menstrual pain (dysmenorrhea), toothache.

Contraindications

  • Hypersensitivity to dexketoprofen or other NSAIDs or to excipients of the drug;
  • a history of bronchial asthma, bronchospasm, acute rhinitis or the development of nasal polyps (allergic-type reactions), urticaria or angioedema when using substances with a similar mechanism of action, such as acetylsalicylic acid and other NSAIDs;
  • peptic ulcer of the stomach or duodenum / bleeding in an active form or a history of recurrence (two or more pronounced episodes of confirmed peptic ulcer, bleeding or perforation);
  • chronic dyspepsia in history;
  • history of gastrointestinal bleeding or perforation associated with the use of NSAIDs;
  • bleeding in the digestive tract, other active bleeding and disorders of the blood coagulation system;
  • Crohn’s disease or ulcerative colitis;
  • severe heart failure;
  • moderate or severe renal impairment (creatinine clearance < 59 ml/min);
  • severe liver dysfunction (Child-Pugh score from 10 to 15 points);
  • hemorrhagic diathesis or other bleeding disorders;
  • known photoallergic or phototoxic reactions during treatment with ketoprofen or fibrates;
  • in patients with severe dehydration (caused by vomiting, diarrhea or insufficient fluid intake);
  • during the third trimester of pregnancy and lactation.
