Pharmacodynamics

Paracetamol has an analgesic and antipyretic effect. Paracetamol blocks cyclooxygenase (COX) I and II only in the central nervous system, affecting the centers of pain and thermoregulation. In inflamed tissues, cellular peroxidases neutralize the effect of paracetamol on COX, which explains the almost complete absence of an anti-inflammatory effect. The lack of influence on the synthesis of prostaglandins in peripheral tissues determines the absence of a negative effect on the water-salt metabolism (sodium and water retention).

Contraindications

  • Hypersensitivity to paracetamol and other components of Anapiron (paracetamol).
  • Severe hepatocellular insufficiency.

Dosage and administration

Anapiron (paracetamol) is used for the rapid removal of pain and / or hyperthermic syndrome, when only the intravenous route of administration of the drug is necessary.

The duration of the infusion should be 15 minutes.

Adults and children weighing 50 kg or more.

The maximum single dose is 1 g of paracetamol, i.e. 1 container (100 ml). The maximum daily dose is 4 g. For patients with an additional risk factor for hepatotoxicity, the maximum daily dose is 3 g. The interval between administration of the drug should be at least 4 hours. Usually, from 1 to 4 infusions are used during the first day from the onset of the pain syndrome (postoperative period), if necessary, the duration of treatment can be increased, but it should not exceed 72 hours (3 days) and a total of 12 infusions.

Adults and children weighing between 33 kg and 50 kg.

15 mg/kg of paracetamol per injection, i.e. 1.5 ml/kg. The maximum daily dose is 60 mg / kg of body weight, but not more than 3 g. The minimum interval between doses should be 4 hours. The duration of treatment usually does not exceed 4 infusions per day.

Children weighing from 10 kg to 33 kg.

15 mg/kg of paracetamol per injection, i.e. 1.5 ml/kg. The maximum daily dose is 60 mg / kg of body weight, but not more than 2 g. The minimum interval between doses should be 4 hours. The duration of treatment usually does not exceed 4 infusions per day.

When using the drug to children, before starting the infusion, the excess of the drug is taken from the container and the volume of the solution is left corresponding to a single dose.

The interval between the administration of the drug in adult patients with renal insufficiency (creatinine clearance £ 30 ml / min) is 6 hours. The duration of treatment should not exceed 48 hours.

For adult patients with hepatocellular insufficiency, chronic alcoholism, chronic malnutrition (low supply of hepatic glutathione), dehydration, the maximum daily dose should not exceed 3 g.
