Itracon Pharmacological properties
Pharmacodynamics. Itraconazole is a triazole derivative. It is active against infections caused by dermatophytes (Trichophyton spp., Microsporum spp., Epidermophyton floccosum), yeasts (Cryptococcus neoformans, Pityrosporum spp., Candida spp., Including C. albicans, C. glabrata and C. krusei), Aspergillus spp., Aspergillus spp. Histoplasma spp., Paracoccidioides brasiliensis, Sporothrix schenckii, Fonsecaea spp., Cladosporium spp., Blastomyces dermatitidis and other varieties of yeasts and fungi. In vitro studies have shown that itraconazole reduces the synthesis of ergosterol, which is an important component of the fungal cell membrane, inhibition of its synthesis provides an antifungal effect.
Pharmacokinetics.
Plasma protein binding of itraconazole is 99.8%.
The therapeutic level in the vaginal tissues remains for 2 days after the termination of the 3-day course of treatment at a dose of 200 mg / day and for 3 days after the completion of the 1-day course of treatment at a dose of 200 mg 2 times a day.
Itraconazole is largely degraded in the liver with the formation of a large number of metabolites.
One of these metabolites is hydroxy-itraconazole, which has an in vitro antifungal effect comparable to that of itraconazole.
About 35% of the dose is excreted in the form of metabolites in the urine for 1 week.
