Pirazinamid Release form and composition
The active ingredient is pyrazinamide, auxiliary components are corn starch, pregelatinized starch, sodium starch glycolate, talc, magnesium stearate, sodium lauryl sulfate.
It is produced in the form of tablets of 500 mg.

Pharmacological properties
A medicinal product intended for the localization and elimination of bacteria of tuberculous activity.
Pirazinamid is active against tuberculosis pathogens that are resistant to other drugs of a similar orientation.
The drug effectively penetrates the core of the tuberculosis lesion. Its effect does not disappear in an acidic environment, therefore it is often used in the treatment of caseous lymphadenitis, tuberculous tumors, pneumonia with the formation of a curdled mass.
Mycobacterium tuberculosis, when monotherapy with Pyrazinamide, develop resistance to the action of the drug, therefore, it is often used in combination with isoniazids and streptomycins.
The drug acts as a bacteriostatic (bactericidal) agent, depending on the dosage and concentration.
Has good absorption and binding to plasma proteins. Begins to act within the first two hours after the initial intake.
In studies, it did not show itself as a carcinogen or mutagen, however, reliable and definitive data on the effect on humans have not been determined.

Contraindications
The drug is not prescribed for impaired liver function and inflammatory processes of the joints.
Caution is prescribed for identified violations in the functioning of the liver, hyperuricemia, hypothyroidism, epilepsy, psychosis, diabetes mellitus.

Pregnancy and breastfeeding period
It can be prescribed during pregnancy, provided that the effectiveness and efficiency of treatment significantly outweighs the risks to the development of the fetus.
Pirazinamid tends to pass into breast milk, therefore, the drug is contraindicated during breastfeeding. However, if there are good reasons to prescribe it to a woman during lactation, then breastfeeding is stopped.

Mode of application
Pirazinamid is taken orally after meals.
The drug is taken 2 tablets twice a day, sometimes the daily dose (2 g) can be divided into three or four doses.
The average dosage for adults is 1.5-2 grams per day. If the drug is well tolerated, then the dosage can be increased to 2.5 g.
For people over 60 years old and children whose body weight is not more than 50 kg, the maximum daily dose is 1.5 g.
The dose is calculated for children based on weight – 20-30 mg is prescribed for each kilogram. The daily dose should not exceed 1.5 grams.

Overdose
Overdose is characterized by the appearance of dysfunction in the liver and a reaction from the central nervous system.
Therapy in case of an overdose is usual – gastric lavage followed by taking sorbents and eliminating symptoms.

Adverse reactions
Therapy is often accompanied by the appearance of allergic reactions – inflammatory skin processes, an increase in the number of eoeinophils in the blood, a sharp increase in body temperature.
It may be accompanied by disorders in the work of the organs of the gastrointestinal tract – nausea, vomiting.
There are headaches, increased nervous irritability, anxiety.
If the course of treatment lasts a long time, toxic liver damage is possible. Therefore, during therapy with Pyrazinamide, it is necessary to observe the state of the liver, to take readings of biochemical samples.
The course of therapy is stopped when changes in the liver appear. To minimize the negative effect on the liver, methionine, glucose, vitamin Bp are prescribed.
Inflammation of the joints and gouty conditions are possible.
