Zoxon 4 (doxazosin) tablets 4 mg. №30

Composition

active substance: doxazosin;

1 tablet contains doxazosin mesylate 2,425 or 4.85 mg, which is equivalent to 2 or 4 mg of doxazosin, respectively;

Excipients: anhydrous lactose, microcrystalline cellulose, granular microcrystalline cellulose, sodium starch glycolate (type A), colloidal anhydrous silica, sodium lauryl sulfate, magnesium stearate.

Dosage form

Tablets.

Basic physical and chemical properties:

Zoxon 2: almost white oblong tablets measuring approximately 9×4.5 mm with a score line and engraved “ZX” and “2” on one side.

Zoxon 4: almost white oblong tablets measuring approximately 12×6 mm with a score line and engraved “ZX” and “4” on one side.

Pharmacotherapeutic group

Antihypertensive drugs. Antiadrenergic drugs with a peripheral mechanism of action. Α-adrenoceptor blockers. ATX code C02C A04.

Pharmacological properties

Doxazosin is a potent and selective antagonist of postsynaptic α1-adrenoceptors. Blocking these receptors leads to a decrease in systemic blood pressure. Drugs Zoxon 2 and Zoxon 4 are intended for oral use once a day in patients with essential hypertension.

Indication

Hypertension

Treatment of hypertension. In most patients, the drug can be used to control blood pressure as monotherapy. If proper blood pressure control cannot be achieved with monotherapy, the drug can be used in combination with antihypertensive agents such as thiazide diuretics, β-adrenoceptor blockers, calcium channel blockers or angiotensin-converting inhibitors.

Benign prostatic hyperplasia

Treatment of urinary tract obstruction as well as symptoms associated with benign prostatic hyperplasia (BPH). To appoint to patients with BPH both in the presence of arterial hypertension, and at normal level of arterial pressure.

Contraindication:

  • hypersensitivity to the active substance or to quinazoline derivatives (eg prazosin, terazosin, doxazosin) or to any of the excipients;
  • cases of orthostatic hypotension in the anamnesis;
  • BPH and concomitant upper urinary tract obstruction, chronic urinary tract infections and the presence of bladder stones;
  • breast-feeding;
  • arterial hypotension in patients with BPH.

Doxazosin as a monotherapy is contraindicated in patients with bladder overflow or anuria with or without progressive renal failure.

Method of application and dosage

The drug can be taken both in the morning and in the evening.
The drug is administered orally.

Hypertension

Treatment should usually be started with Zoxon 2, which contains less active substance. Take the drug once a day: the initial dose is 1 mg (1/2 tablet of Zoxon 2) for 1-2 weeks to minimize the risk of orthostatic hypotension and / or syncope. After 1-2 weeks, the dose can be increased to 2 mg (1 tablet of Zoxon 2) per day, if necessary. In case of insufficient effectiveness, the daily dose can be gradually increased taking into account the individual patient’s response to treatment, which is assessed depending on the achievement of the desired level of blood pressure.

In most cases, use a dose of 2-4 mg per day (1 tablet of Zoxon 2 or 1 tablet of Zoxon 4), which should be taken once a day. If necessary, the dose can be increased to 8 mg or to the maximum recommended dose of 16 mg.

Benign prostatic hyperplasia.

Treatment should usually be started with Zoxon 2, which contains less active substance. The recommended starting dose is 1 mg (1/2 tablet) to be taken once a day to minimize the risk of orthostatic hypotension and / or syncope.

Depending on the individual characteristics of the patient’s urodynamics and symptoms of BPH, the dose can be further increased to 2 mg (1 tablet of Zoxon 2), then to 4 mg (1 tablet of Zoxon 4) and to the maximum recommended dose of 8 mg. The recommended dose titration interval is 1-2 weeks. The usual recommended dose is 2-4 mg per day (1 tablet of Zoxon 2 or 1 tablet of Zoxon 4), which should be taken once a day.

Overdose

The main symptom of doxazosin overdose is hypotension. The patient should be placed on his back immediately with his head down. Treatment is symptomatic, aimed at stabilizing hemodynamic parameters and vital functions of the body. If symptomatic measures are insufficient, plasma substitutes should be used first to treat shock. After that, if necessary, use vasoconstrictors. Renal function should be monitored and supportive measures instituted if necessary. Due to the high degree of plasma protein binding of doxazosin, hemodialysis is not indicated.

Side effects:

  • infections and invasions: Respiratory tract infections, urinary tract infections;
  • from the nervous system: drowsiness, dizziness, headache;
  • from the organs of hearing and vestibular apparatus: vertigo;
  • from the heart: increased heart rate, tachycardia
  • from vessels: arterial hypotension, orthostatic arterial hypotension;
  • from the gastrointestinal tract: abdominal pain, dyspepsia, dry mouth, nausea.
