Pharmacological properties
Pharmacodynamics.
Indomethacin is an indoleacetic acid derivative belonging to the group of non-steroidal anti-inflammatory drugs. Indomethacin has a powerful inhibitory effect on prostaglandin synthesis by suppressing the enzyme cyclooxygenase, which plays a significant role in reducing inflammation, pain and hyperthermia. It has antipyretic properties and reversibly inhibits platelet aggregation. When applied topically, indomethacin has a direct anti-inflammatory and analgesic effect on inflamed tissues.
Pharmacokinetics.
The ointment is well resorbed through the skin and accumulates in subcutaneous tissues at therapeutic concentrations, including joints and synovial fluid. It is excreted by the kidneys in the form of metabolites and in small quantities – unchanged, including when applied topically; in small amounts it is excreted through the biliary tract into the intestines.
