Description

Urorek (silodosin) hard capsules 4 mg. №30

Composition

active substance: 1 capsule contains 4 mg or 8 mg of silodosin;

Excipients: mannitol (E 421), peppeted corn starch, purified water, sodium lauryl sulfate, magnesium stearate, gelatin, titanium dioxide (E 171), for a dosage of 4 mg of yellow iron oxide (E 172).

Dosage form

Hard capsules.

Basic physical and chemical properties:

hard capsules containing 4 mg of silodosin,

hard capsules containing 8 mg of silodosin.

Α-adrenoceptor antagonists. Silodosin.

Pharmachologic effect

Highly selective competitive antagonist of α1A-adrenergic receptors, which are mainly located in the prostate gland, in the tissues of the fundus and neck of the bladder, the capsule of the prostate gland and the prostatic part of the urethra.
Blockade of α1A-adrenergic receptors reduces the tone of smooth muscles in these tissues, which leads to an improvement in the outflow of urine from the bladder. At the same time, the symptoms of obstruction and irritation associated with benign prostatic hyperplasia are reduced.
In vitro studies have shown that the affinity of silodosin for α1A-adrenergic receptors is 162 times greater than its ability to interact with α1B-adrenergic receptors, which are located in vascular smooth muscles. Due to its high selectivity, silodosin does not cause a clinically significant decrease in blood pressure in patients with initially normal blood pressure.

Indications of the drug Urorek

Treatment of symptoms of benign prostatic hyperplasia.

Contraindication

Hypersensitivity to silodosin or any of the excipients in the drug.

Method of application and dosage

Adults. Apply internally. The recommended dose is 8 mg once a day. For certain groups of patients, 1 capsule of Urorek 4 mg once daily is recommended (see below).

The drug Urorek hard capsules should be taken with food, preferably at the same time of day. The Urorek hard capsules should not be broken, it should be swallowed whole, not chewed, washed down with a glass of water.

Children

Do not use in pediatric practice.

Overdose

Silodosin was evaluated at doses up to 48 mg / day taken by healthy male volunteers. Orthostatic hypotension was a dose-limiting adverse reaction.

If the Urorek hard capsules has been taken recently, it is recommended to induce vomiting or gastric lavage. If an overdose of Urorek is accompanied by hypotension, it is necessary to provide the patient with support of the cardiovascular system. Dialysis is impractical because silodosin in the body is almost completely bound to blood proteins (96.6%).

Side effects:

  • from the nervous system: dizziness;
  • from the vascular system: orthostatic hypotension;
  • respiratory, thoracic and mediastinal disorders: nasal congestion;
  • from the gastrointestinal tract: diarrhea;
  • from the reproductive system and mammary glands: retrograde ejaculation, anejaculation.
